Production of α-Amylase from Bacillus megaterium MD-1

The alpha-amylase is used extensively in many different industrial sectors and renowned for modifying starch by rupturing 1-4 glycosidic bands. Depending on the intrinsic properties of microorganism, several alpha-amylases with thermostable halotolerant are expressed. In current study, bacteria were isolated from Ergani Makam Mountain. Identification optimization performed. As a result 16S rRNA analysis, physiological, morphological biochemical analyis carried out identification microorganism consequently bacterium was defined as Bacillus megaterium MD-1.
 Following its identification, α-Amylase, B. megaterium. Optimal conditions enzyme production determined 48 hours, 35°C pH 7.0. Maximum activity optained at 40°C 8.0. effects various carbon nitrogen sources investigated adding to nutrient medium. Compared control regarding production, it that sources, particularly sucrose, fructose lactose inhibited 75%, no change other hand observed glucose, galactose. It also urea sodium nitrate had an inhibitory effect whereas did not. highest amylase among obtained peptone addition.
 our increase could be achieved using optimum values physical parameters.
